Is Bottled Water Worth It? Essay

In somewhat recent discussions, one of the worlds favorite grab and go drink has come into question whether it’s safe enough to drink or not and that is, bottled water. A well-known convenient commodity to everybody, but some argue it is neither healthy for our environment, our pockets, or ourselves. The people from this perspective say bottled water deserves to be banned because the negatives indeed out weigh the positives. On a different side of the spectrum others use bottled water’s good marketability, convenience and it being a better alternative to soda and other sugary drink found in schools as excuses to keep bottled from being banned. Another interesting view expresses the fact that bottled water is silly in itself but it still should not be banned. So the question is to be banned or not to be banned? Millions of people drink it a day but do they ever question whether what may be in the water they’re drinking? Probably not, nor should they have to. The Environmental Working Group (EWG), a nonprofit organization that strives to protect the public health and the environment using public information (Langwith 1), had one of the United States leading water quality laboratories test 10 commonly bought bottled water brands in 9 states and in the District of Columbia. What they found might be astonishing to some. They found these brands tested contained a total of 38 chemical pollutants and an average of 8 contaminants in each brand. A third of the chemicals found were not even regulated in bottled water (Langwith 2). Some of the chemicals and pollutants that were found in these 10 brands were things from disinfection products to caffeine and pharmaceuticals. Heavy metals and minerals like arsenic and radioactive isotopes were also found (Langwith 2). It is also not mandatory for bottled water companies to inform their customers when they get contaminants in their water (Langwith 3). So essentially, you don’t know exactly what your getting when decides to drink bottled water. Besides the effects it has on our health it also wastes our money. Two of the brands that were tested, Wal-Mart’s Sam’s Choice and Giant Acadia, were found to be chemically indistinguishable from tap water (Langwith 3). Basically people are just buying they’re water back for 1,900 the price of the tap water they could be getting at their homes for much cheaper. Damaging to the environment as well, out of the 36 billion bottles sold in 2006 only a fifth of them were recycled. The rest of the bottles ended up as litter in places such as landfills, land and in rivers, streams, and oceans. Producing water bottles in the U.S. uses around 1.5 million barrels of oil every year. This is enough energy to power 250,000 homes or fuel for 100,000( Langwith 5). Taking water for plastic bottles puts quite a strain on our rivers, streams, and community drinking water supplies. When water isn’t bottled from a municipal supply these companies must draw it from groundwater supplies, rivers, springs or streams (Langwith 5). Some people view bottled water as a good thing from a marketing point of view and a way to help children in general and in schools. With obesity rates rising in the United States claiming that 17 percent of children in the United States are overweight. Bottled water could be used as a healthier alternative then all the sugary sodas and other drinks that are found in schools. Schools and companies are trying to make bottled water a fun and healthy. Nestle, a bottled water manufacturer came up with the Aquapod, which is a rocket ship shaped bottle to attract the younger buyers (Tucker, Sprague 3). Other brands have also made fun water bottles as well. Bottled water is a constantly growing business. During 2007, bottled water accounted for 17 percent of the non-alcoholic beverages. By the year 2011 the bottled water market grew about 48.5 percent (Tucker, Sprague 3). Charles Fisherman provides an interesting view to the subject stating that bottled water is silly, yet it shouldn’t be banned. He gives an example of the silliness of water bottles with the company Pellegrino and how they wash their bottles with Pellegrino water. But he also says banning water bottles wont teach anyone anything. Regardless there will still be soda and other drinks available that are high in sugar for people to consume. People also need bottled water on campuses because they’re water fountains are dirty and unsafe to use (Fisherman 2). Though these arguments give valid points but do not address the effects that bottled water may have on ones health and environment that in turn is more important then its marketability and how convenient it may be.

Life Ambitions Essay

Owen Stuart Phillips-Statement of Ambitions and Life Purpose Scouting has taught me many things about myself. It is a big part of who I am now and it will stay with me forever. Through Scouting, I have learned the importance of preparation, personal responsibility, service to others, and leadership skills. I first started out with Cub Scouts where I earned every rank and received my Arrow of Light. After Cub Scouts, I crossed over to Boy Scouts and joined Troop 40. I have been extremely involved with my Troop and I have done almost everything they have had to offer. I am proud of the fact that I will be the 6th member of my extended family (cousins) to earn the rank of Eagle Scout from Troop 40, all under the guidance of the same Scoutmaster, Mr. Clifton Glover. I plan on staying involved Scouts to improve on my leadership skills and earn additional merit badges. I would also like to earn the three Eagle Palms. In school I am an honor roll student. I am in the band and I have received awards at band competitions in Greenville at ECU and Music Performance Adjudication held in Jacksonville. When I was in the 5th grade I completed the DARE program which stands for Drug Abuse Resistance Education. We were required to write a 2 page essay to complete the course. The winning essay would be read aloud by the student at the graduation ceremony. My essay won the competition, I received a trophy and I was very proud. I play sports such as football, baseball, and wrestling. I have been team captain in football. I was also asked to participate in a youth football instructional video for Championship Productions. The video was filmed at Wingate University with other youth players and college coaches, demonstrating football drills and fundamentals. The video will be sold to youth players and coaches nationwide. After I graduate High School, I have strong interests in applying to our Military Service Academies. I would like to be an officer in the Military because I feel like I can be a leader instead of a follower. I started getting strong interests in the Military Academies when we went on a camp out to the United States Military Academy at West Point. It was a great experience and I absolutely loved every bit of it. If it was not for Scouting, I would have not had that opportunity. I enjoy math and engineering, and would like to pursue a career in this area in the military, such as a combat engineer.

This thickened mesenteric fat y extends up to the serosal surface of the bowel causing Mr. Jackson gastrointestinal tract to appear mildly obese.   Pain in the left lower quadrant of the gastrointestinal tract again shows signs and the presence of the inflammation. Moreover, it is the small intestine from where the nutrients are absorbed in the blood stream. High level of inflammation followed by formation of the granuloma in the small intestine prevents this normal absorption of the nutrients and thus further generating the pain. Furthermore, this inflammation in the gastro-intestinal tract leads to the generation of the delayed type hypersensitivity response or other type I and Type II hypersensitivity response. This rise in the hypersensitivity response leads to the increase in the levels of the cytokines mostly IL-8. High levels of IL-8 in the blood caused problem with the urinalysis (Steenholdt et al., 2012).    Headache in the last few days as encountered by Mr. Jackson may not have a direct relation with the pathophysiology of the Crohn’s disease however, increase in the level of abdominal pain, lack of nutrient and lack of sleep might be the reason behind this sudden headache. Thus from the above detailed analysis of the condition of Mr.Jackson on the basis of disease aetiology and pathophysiology, it seems that Mr. Jackson might be suffering from Crohn’s disease The symptoms of crohn’s disease and Ulcerative Colitis are more or similar. The common symptoms of the Crohn’s disease and Ulcerative Colitis are: So taking the medical condition and the symptoms Mr. Jackson into consideration, he becomes a bit dubious to claim that he is suffering from Crohn’s disease. He might also be the victim of the Ulcerative Colitis. Ulcerative colitis affects colon and the rectum leading to chronic inflammation. Thus, Ulcerative colitis affects mostly the lower portion of the gastrointestinal whereas; Corhn’s disease can affect or may cause inflammation to any portion of the gastrointestinal tract (Bressler et al., 2015). In order to detect the exact Inflammatory Bowel disease that has affected Mr. Jackson, a ultrasonography of the entire stomach is mandatory.   The ultrasonography will reveal the exact point of inflammation via show casing the organo-megally. It is in the ileum or in the upper portion of the respiratory tract then it will be Crohn’s disease while if the organ enlargement is seen in the rectum or colon then Ulcerative Colitis can be confirmed (Chatu, Subramanian & Pollok, 2012). Crohn’s disease as discussed is caused by Mycobacterium avium paratuberculosis whereas Ulcerative Colitis is mostly caused by Escherichia coli (Winter et al., 2013). A bacterial culture test using the selective media for the Escherichia coli and Mycobacterium avium paratuberculosis will help in the elucidation of the exact micro-organism colonizing in the gastrointestinal tract leading to the elucidation of the disease. TIRF microscopy is one method to count molecules. The bacterial flagellar motor is a complex system in which motility protein B molecules can be counted using TIRF. Introduction: The bacterial flagellar motor, TIRF microscopy and associated research Flagellar motors are machines used to drive many bacteria which have to swim in a solution like our bodies. This motor, usually studied in E. coli bacteria, is powered by a flux of H+ or Na+ ions across a cytoplasmic membrane driven by an electrochemical gradient (Sowa and Berry, 2008). The motor itself consists of two components, a rotor and a stator: the rotor spins relative to the cell and is attached by a helical filament known as a hook, whereas the stator is fixed to the cell wall (Francis et al 1994). A method commonly used to visualise the bacterial flagellar motor is Total Internal Reflection Fluorescence (TIRF) microscopy, which is one of the most frequently employed methods in bio-optical research (Leake 2013, P87). TIRF microscopy uses an evanescent field to illuminate the area covered by the specimen in question, which is adjacent to a glass-water interface. Using organic dyes has made it possible to view other properties of bacteria using TIRF (Sako et al 2000). This method is useful in counting the molecular subunits of the bacterial flagellar motor. TIRF microscopy has been used to view single molecules within live bacteria. For viewing the bacterial flagellar motor of E. coli, scientists tagged motility protein B (MotB) cells with Green Fluorescence Protein (GFP) in order to detect them via TIRF. This highlighted the areas within the bacteria where the motor was situated. To visualise the bacteria in a single confined position, the cell was tethered to the slide for viewing on the microscope. This is shown in Fig. 1, where the fixed position of the flagellum limits the bacteria’s movement to rotation. Background, difficulties and discoveries from the research 2.1 The history behind counting molecules Though the basis of this experiment began in the 60s, initially using the measurement of the activity of single molecules (Rotman 1961), optical detection and spectroscopic methods are now used instead. The counting of complex molecules can now also be achieved, but this area of research also needs TIRF microscopy. TIRF was enhanced in 1984 by Daniel Axelrod after the publishing of a paper on its experimental methods (Axelrod 1984), and those methods remain largely unchanged today. Furthermore, GFP molecules have only been recently understood. Without this research and development in GFP, visuals using TIRF would not be possible (Tsien 1998). 2.2 The difficulties encountered and overcome in counting molecules An estimate of around twenty-two molecules are thought to be present in the flagellar motor, with roughly eleven stator units. The main issue with determining this result explicitly is that there are many MotB molecules not associated with the motor. These molecules cause a problem as they are free to diffuse within the motors of the cell membrane. The fluorescence intensity was estimated from the areas where it was clear that no such molecules would interfere with results. Additionally, an intense laser beam focus for TIRF was required to photobleach GFP molecules. Only an extremely small region of the bacteria was viewed to improve the ability to track a small number of molecules – a significant amount of noise remained in the system, however, meaning that it is not yet possible to count exactly how many molecules are in each motor. Using TIRF, bright spots indicate the centre of the cell rotation of the image shown in Fig. 2. There was a high density of spots centred on the flagellar motor, due to the high density of GFP-MotB molecules around the motor. Short times (between 0-10 seconds) are used because TIRF illumination over the bright spots decreases over longer periods of time, which makes it difficult to detect regions of the flagellar motor. Care was taken to not cause damage to the GFP due to the excitation light on the surrounding water: this means that smaller time steps were required such that the GFP molecules emitted a constant amount of photons. 2.3 The effects FRAP and FLIP With the noise effects reduced, there was the opportunity for the GFP-MotB molecules to spread into the area which had been bleached beforehand. Focusing the laser beam onto the motor itself resulted in the effects of fluorescence recovery after photobleaching (FRAP) and fluorescence lost in photobleaching (FLIP). Observations of the molecular turnover in the cell found that over a period of five minutes, the intensity of the bright spots around the motor would decrease to nearly zero but then recover to half the initial intensity. This implied that binding and unbinding at the motor and bleaching occurred in the evanescent field (Leake et al 2006, P357), which means that the stator units in the motor only spend half a minute in each flagellar motor. This is demonstrated in Fig. 3, which illustrates the time elapsed after laser focused bleaching and how the intensity decreases but eventually recovers. Fig. 3. Benefits of the research and potential uses for the future An improved type of MotB was used in the research, which enhanced the way in which the molecules in the motor were counted. FLIP and FRAP indicate an alternative means for visualising the motor in motion, confirming that the stator units are dynamic instead of static (Sowa and Berry, 2008, P117). This is one of the first measurements of turnover in a molecular machine, establishing other possible characteristics which could be exploited to gain further understanding of the motor (Leake et al 2006, P357). Scientists are keen to understand more about how such motors work, so that developments in the delivery of medicine or for environmental purposes can be made. It may be possible to replicate the motor (Fukuda et al 2012). Delivery of medicine is one of the key goals: modelling the bacterial flagellar motor such that it could be used for targeted drug delivery would be revolutionary (Leake 2013, P259). Furthermore, through the development of nano-bots, this could be used to visualise diseased tissue or uncover parts of the human body. Summary This area of biophysics is relatively new: from the discoveries in the early 60s through to the 80s, there has not been a clear link between the two subjects. From the late 90s there was an opportunity to visualise biological material using physical optical devices. Over the past two decades, it has now reached to the point where it is possible to count single molecules to a close estimate. The use of GFP molecules combined with TIRF can enhance the visualisation of molecules in bacteria, and there are methods which can significantly improve the estimation of the number of molecules in the motor.
